Passion Flower Extract aids deep sleep through its interaction with GABA receptors in the central nervous system. Passion flower has also shown anxiolytic (anti-anxiety) properties in some studies, which may be due to its GABAergic effects as well as other mechanisms. Reducing anxiety and inducing a state of calm can help prepare the body for sleep. Some compounds in passion flower, such as flavonoids and alkaloids, have been shown to have mild sedative effects. Mucuna pruriens is a tropical legume that has been used traditionally for various medicinal purposes It is a natural source of L-DOPA, a precursor to the neurotransmitter dopamine. Mucuna pruriens, through its L-DOPA content, can convert to dopamine in the brain. Dopamine plays a complex role in sleep regulation, including the promotion of REM sleep. Corydalis yanhusuo is a traditional herbal remedy commonly used in Traditional Chinese Medicine (TCM). One of the primary ways Corydalis yanhusuo is believed to contribute to falling asleep faster/ sedation is through its interaction with the GABA (gamma-aminobutyric acid) system where it inhibits the reuptake, thereby increasing the availability of GABA in the synaptic cleft.

GABA is an inhibitory neurotransmitter, which means that it reduces neuronal excitability. 5-HTP is a naturally occurring amino acid derivative. It is produced in the body from the amino acid tryptophan.

Unlike other precursors like tryptophan, 5-HTP bypasses the rate-limiting step of serotonin production (conversion of tryptophan to 5-HTP via the enzyme tryptophan hydroxylase) and can readily cross the blood-brain barrier. This makes it more effective at increasing serotonin levels.

The primary physiological role of 5-HTP is as an intermediate in the synthesis of the neurotransmitter serotonin. The pathway is as follows:

  1. Dietary tryptophan is converted into 5-HTP via the enzyme tryptophan hydroxylase. or 5-HTP is supplemented.
  2. 5-HTP is then decarboxylated by the enzyme aromatic L-amino acid decarboxylase to produce serotonin.

Given its role in serotonin synthesis, 5-HTP is often used therapeutically, primarily in supplement form, for conditions associated with low serotonin levels, such as:

Thermogenesis is the process by which your body generates heat by burning calories. The calories burned either come from immediate food intake or stored body fat. There are 3 types of thermogenesis: Basal Metabolic Rate (BMR) or Resting Metabolic Rate (RMR) which is the number of calories burned at rest, Activity-Induced Thermogenesis (AIT) which is energy expended during activity/ exercise, and Diet-Induced Thermogenesis (DIT) which refers to the energy expended during digestion, absorption and distribution of nutrients. However, imbalanced lipid levels, such as high levels of LDL ("bad" cholesterol) and triglycerides or low levels of HDL ("good" cholesterol), can contribute to metabolic disorders like obesity, diabetes, and cardiovascular diseases

Brown Adipose Tissue and Fat Oxidation

Brown Adipose Tissue (BAT), is one of two types of fat found around the body, the other being White Adipose Tissue (WAT)/ Stubborn Soft Fat. Unlike WAT, BAT is known for its ability to invoke Thermogenesis and rather than storing fat, burns it. BAT is packed with mitochondria, which are the powerhouses of cells that convert fatty acids into energy, causing thermogenesis. Within these Mitochondria there is a unique protein called Uncoupling protein 1 (UCP1), this protein is largely responsible for the thermogenic properties of BAT. Usually, when fatty acids are oxidised in the mitochondria, the energy released is used to pump protons across the mitochondrial membrane, creating a gradient. This gradient is then used to make Adenosine Tri-phosphate (ATP), the body's main form of energy. UCP1 disrupts this process and allows the protons to flow back across the membrane without making ATP, causing the energy to be released as heat. A study published to the British journal of nutrition in 2013 was first to report that Paradoxine (Grains of Paradise) supplementation leads to greater energy expenditure and brown fat stimulation compared to the placebo group. BAT activation leads to greater weight loss but also can turn WAT into BAT, this process is called "browning".

Effectively, Grains of Paradise increases BAT activation, the amount of browning that occurs within your body and also increases Fat Oxidation. 

Fat oxidation is the process which stored fatty acids are broken down and converted into usable energy, this process occurs in the mitochondria of cells. The first step to fat oxidation is lipolysis, where stored fatty acids are broken down into free fatty acids. this process is regulated by hormones such as epinephrine, growth hormone, and glucagon. Once released into the blood stream, they bind to carrier proteins called albumin and are then transported to various areas around the body such as: skeletal muscle, liver and heart, where they can be utilised as energy. Beta-Oxidation then occurs when once the fatty acids are transported through the cell into mitochondria. the Mitochondria then break down the fatty acids into two carbon units known as acetyl-COA which then enter the citric cycle (Krebs Cycle). In this cycle they are further oxidised, leading to production of Adenosine Tri-Phosphate (ATP).
